An improved clearing and mounting solution to replace chloral hydrate in microscopic applications1

چکیده
UNLABELLED PREMISE OF THE STUDY This study presents Visikol™, a new proprietary formulation that can be used as an efficient replacement for chloral hydrate as a clearing agent for microscopic examination. In the United States, chloral hydrate is regulated and therefore difficult to acquire. • METHODS AND RESULTS Fresh and dry samples of the following plants: ginger (Zingiber officinale), maté (Ilex paraguariensis), lime basil (Ocimum americanum), oregano (Origanum vulgare), and mouse-ear cress (Arabidopsis thaliana), were cleared using Visikol or chloral hydrate solution and compared using a light microscope. • CONCLUSIONS This new method can be used successfully to clear specimens, allowing identification of diagnostic characteristics for the identification of plant materials. Visikol is as effective as chloral hydrate in providing clarity and resolution of all tissues examined. Tissues become transparent, allowing observation of deeper layers of cells and making it effective in research, botanical and quality control, and for educational applications.

A clearing protocol for whole tissues: An example using haustoria of Orobanchaceae1
UNLABELLED PREMISE OF THE STUDY Due to lack of success in clearing whole tissues using only classical clearing techniques (e.g., Herr's 4½ solution, KOH, NaOH, lactic acid saturated with chloral hydrate), and because tissue degradation is often a result of harsh clearing agents (e.g., KOH, NaOH), a novel combined treatment was sought to improve the removal of obscuring tannins from intact ha...
متن کاملComparison effect of pentobarbital sodium with chloral hydrate anesthesia on post-ischemic damage in an experimental model of focal cerebral ischemia
Introduction: Anesthetic agents, blood pressure, arterial pH and blood gases have found to influence on the pathophysiology of experimental stroke. Despite, there are very few comparative studies about effects of anesthetic agents in animal model of cerebral ischemia. Therefore, in this study, we investigated the effects of chloral hydrate and pentobarbital anesthesia, as comparative study, on...
متن کاملAn improved method for clearing and staining free-hand sections and whole-mount samples.
BACKGROUND AND AIMS Free-hand sectioning of living plant tissues allows fast microscopic observation of internal structures. The aim of this study was to improve the quality of preparations from roots with suberized cell walls. A whole-mount procedure that enables visualization of exo- and endodermal cells along the root axis was also established. METHODS Free-hand sections were cleared with ...
متن کاملCombination of Lidocaine and Chloral Hydrate as a New Method of Anesthesia in a Rat Model of Ischemic Stroke
Background and purpose: In experimental studies, proper choice of anesthesia method is important for both validity of results and improving the rate of success and mortality. In current study, we examined the effects of lidocaine combined with chloral hydrate for induction of anesthesia in a rat model of ischemic stroke. Materials and methods: In this research, 30 male rats were studied as sha...
متن کاملProtective effect of chloral hydrate against lipopolysaccharide/D-galactosamine-induced acute lethal liver injury and zymosan-induced peritonitis in mice.
In recent years, certain anesthetics have been shown to have protective effects against acute inflammation in experimental animals, an observation that may yield new options for adjunctive treatment of acute inflammation. In this study, we investigated the effects of chloral hydrate (CH) on the acute inflammatory response in BALB/c mice using lipopolysaccharide/D-galactosamine (LPS/D-GalN)-indu...
